| Summary: | Coffee culture has various sanitary problems, among them, showed up to order fungal diseases, such as blister spot, caused by Colletotrichum gloesporioides Penz, which has deteriorated in areas producing coffee (Coffea arabica L.). The lack of information about the Colletotrichum pathosystem x Coffee difficult to elucidate aspects of the pathogenicity of races and transmissibility. The use of genetically modified microorganisms using molecular markers has been an important tool for these types of studies. Thus our study aimed to evaluate the efficiency of two systems of genetic transformation using the marker gene gfp (Green Fluorescent Protein), as well as cultural characteristics, stability and processed, and pathogenicity of isolates of Colletotrichum gloeosporioides processed. It found success using the two processing systems, confirmed this by means of epifluorescence microscopy and resistance to the antibiotic hygromycin-B, where incorporated into the culture medium. The fungus maintained its cultural and morphological characteristics compared to the wild strains. To be inoculated on seedlings of coffee can be seen that the pathogenicity of the isolates processed were not changed after the transformation. |
